What is claimed is:     1. A substantially pure polypeptide comprising a mutant diphtheria toxin R domain, said R domain comprising a mutation in at least one or more residues from the group consisting of Lys 516, Lys 526, Phe 530, and Lys 534 of wild-type diphtheria toxin, said polypeptide binds sensitive cells with less affinity than wild-type diphtheria toxin and forms an immune complex with an antibody which specifically recognizes the R domain of wild-type diphtheria toxin.
